H9 human embryonic stem cells (hESCs) were treated with an adeno-associated virus (AAV) vector carrying a green fluorescent protein transgene cassette driven by a CMV promoter. Cells treated with vector possessing an AAV serotype 3 capsid demonstrated toxicity, whereas cells treated with a vector possessing an AAV serotype 4 capsid demonstrated no toxicity (see control video; this lack of toxicity is believed to be due to AAV4 not uncoating in these cells).
